T reg cells are commonly thought to be generated in the thymus because of recognition of self-antigens ( 3, 4 ), which is supported by studies using mice with limited TCR diversity showing strong similarity between the thymic and peripheral T reg TCR repertoires ( 5 – 7 ). However, the peripheral and thymic T reg TCR repertoires were not identical, suggesting that certain TCRs are preferentially enriched or deleted in the periphery, akin to earlier observations in the total CD4 +

Antigen-specific peripheral shaping of the natural regulatory T cell population

Although regulatory T (T reg) cells are thought to develop primarily in the thymus, the peripheral events that shape the protective T reg cell population are unclear. We analyzed the peripheral CD4(+) T cell receptor (TCR) repertoire by cellular phenotype and location in mice with a fixed TCRbeta chain. We found that T reg (Foxp3(+)) cells showed a marked skewing of TCR usage by anatomical loca...

Expansion of CD4+CD25+FoxP3+ Regulatory T Cells in Chronic Hepatitis C Virus Infection

Background: Regulatory T cells (Tregs) have been involved in impaired immunity and may have a pivotal role in persistence of viral infections. Objective: To develop a simple and reliable in-house three color flow cytometery of peripheral blood to understand the role of HCV infection in the increase of Tregs. Methods: The level of naturally occurring CD4+CD25+FoxP3+ regulatory T cells (nTregs) i...

P-212: Association between Polymorphisms of CTLA-4 Gene and Unexplained Recurrent Spontaneous Abortion in An Iranian Population

Background: Unexplained recurrent spontaneous abortion (URSA) has been suggested being associated with the failure of fetal-maternal immunologic tolerance in which the regulatory T lymphocytes (Tregs) play a crucial role. Cytotoxic T lymphocyte-associated antigen 4 (CTLA-4) is expressed transiently on activated T cells and constitutively on Tregs. CTLA-4 interacts with the B7 cell surface molec...

SUSCEPTIBILITY OF HUMAN WM MELANOMA CELL LINES TO NK AND LAK CYTOTOXICITY AND THEIR RELEVANCE TO THE LEVEL OF MHC CLASS I AND ICAM-l ANTIGEN EXPRESSION

The effect of natural killer (NK) cells and lymphokine activated killer ( LAK) cells was studied on a group of human melanoma cell lines. Peripheral blood from healthy volunteers was utilized as a fresh source of natural killer cells and rhI L-2 for producing LAK cells. The cytotoxicity of effector cells was quantified using a 4 hour SI determining the density of antigen expression on tumor...

FoxP3+ Regulatory T Cells in Peripheral Blood of Patients with Epithelial Ovarian Cancer

Background: Ovarian cancer is the fifth leading cause of death from malignancy in women. CD4 +CD25+FoxP3+ regulatory T (Treg) cells are a subset of T lymphocytes with great inhibitory impact on immune response. Objectives: To investigate the percentage of CD4 +CD25+FoxP3+ regulatory T cells in the peripheral blood of the Iranian patients with epithelial ovarian cancer compared to healthy women ...
